PDRN serums — what the salmon-DNA evidence actually shows
Almost all of the strong clinical evidence for PDRN comes from injections, not creams. What the topical evidence covers so far, and how to read a salmon-DNA serum honestly.
Almost all of the strong clinical evidence for PDRN comes from injections, not from creams. That is the single most useful fact to hold when reading a salmon-DNA serum's label — the ingredient is real and well studied, but nearly all of the studies bypass the barrier that a serum has to cross.
This is not a reason to dismiss it. It is a reason to ask a different question than the one the marketing asks.
What is PDRN, and why is it called salmon DNA?
PDRN stands for polydeoxyribonucleotide: a mixture of DNA fragments, usually purified from salmon or trout sperm. In aesthetic clinics the closely related term is polynucleotide, and the two are often used interchangeably in marketing even though the preparations differ.
What matters cosmetically is that these are long molecules, not small actives — the fragments run to hundreds of kilodaltons, thousands of times larger than a typical skincare ingredient. Size is the whole story of what follows.
Does PDRN work when you apply it to the skin?
The honest answer is that the evidence base is thin and very new, while the injectable evidence is larger and older.
A systematic review of polynucleotides in aesthetic medicine pooled nine studies covering 219 patients, all using intradermal injection. It found promising results for wrinkles, texture and elasticity, but rated the included studies as low to moderate quality and concluded that rigorous trials are still needed (Lampridou et al., 2025).
For topical use, the frequently cited laboratory work does not test what a serum actually does: one widely referenced study applied PDRN together with vitamin C and niacinamide to mice, delivered through microneedling — a mixture, an animal, and a route that punctures the barrier rather than crossing it (Kim et al., 2022).
There is a mechanical reason for the gap. As a rule of thumb, compounds above roughly 500 daltons do not passively cross intact stratum corneum (Bos & Meinardi, 2000). PDRN sits far above that line.
Is a PDRN serum better than retinol?
One recent study makes that claim, and it deserves to be read carefully rather than repeated. In a randomised, double-blind, split-face trial, 31 women aged 35 to 55 used an eye cream containing a medium-length PDRN preparation on one side and 0.1% retinol in an identical base on the other, twice daily for 28 days. The PDRN side showed roughly twice the improvement in periocular wrinkles and dermal density (Ye et al., 2026).
That is a genuinely promising result, and the design is better than most cosmetic testing. But note the clock. Twenty-eight days is far too short for retinol to show what retinol does — the trial that established retinol's effect on aged skin ran for 24 weeks — so outperforming retinol at four weeks is not the same as outperforming retinol.
It is one study, on one small group, on the eye area only, and it has not yet been replicated. If you are choosing an ingredient to commit to for a year, the one with decades of trials behind it is still the safer bet.
Simply put: judge the route, not the ingredient list
An ingredient list tells you what is in the bottle. It cannot tell you what reaches the layer where the ingredient would have to act. For a molecule this large, that gap is not a detail — it is the entire question.
So the practical way to read a PDRN serum is to assume its measurable benefit comes mostly from its base: the humectants, lipids and soothing agents carrying it. That is not a criticism. A well-built base is genuinely useful, and skin that is holding water and calm is doing most of what these products promise. It just means you should be paying for the formula, not the headline.
How to read a PDRN product before you buy
- Check where PDRN sits in the ingredient list. If it appears after the preservatives, it is present at a token level and the base is doing the work — buy it for the base or not at all.
- Ask what evidence the brand is citing. If the study is an injection trial, a cell assay or a mouse model, it is not evidence about the cream in your hand.
- Treat it as an addition, not a replacement. Keep the barrier work and daily sun protection in place, and judge any new serum after eight weeks, not eight days.
- If your interest is the eye area specifically, compare it honestly against the options that already have longer records there before adding another step to a routine that may already be full.
